title | | Reduction of overflow metabolism in E. coli by increasing the respiratory capacity |
abstract | | Aerobic acetate production (known as overflow metabolism) remains as a serious challenge for reaching high cell-densities and fully exploiting the biosynthetic potential of E. coli. While most process and metabolic approaches to avoid overflow metabolism have been designed to limit the glucose uptake rate, it can result in a reduction of growth rate. Instead of reducing glucose uptake, our research group has increased the respiratory capacity of E. coli. By aerobically expressing the Vitreoscilla hemoglobin, higher glucose uptakes were obtained, but also faster growth rates and a strongly reduced acetate accumulation. Data of batch cultivations as well as the additional effect of arcA mutation will be shown.
